UASt regulatory sequences drive expression of a blocker of nuclear Ca[2+] signaling. This consists of four copies of the M13 peptide from Ggal\MLCK, each of which contains an N-terminal cryptic nuclear localization signal which is sufficient for nuclear localisation. In addition, a Tag:MYC epitope has been inserted at the C-terminal end of the coding sequence.
Flies expressing Ggal\MLCKCaMBP4.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4hs.PU during conditioning (expression induced by a 30 minute heat shock 12-18 hours prior to conditioning) show no significant impairment in short-term memory, middle-term memory or anesthesia-resistant memory, but show a significant impairment in long-term memory, in an olfactory avoidance conditioning assay.
Flies expressing Ggal\MLCKCaMBP4.Scer\UAS.T:Hsap\MYC under the control of either Scer\GAL4ey-OK107 or Scer\GAL4Tab2-201Y for 5 to 6 days prior to testing (using the temperature sensitive Scer\GAL80ts.αTub84B allele to prevent expression during development) do not show defects in short-term memory, but show a significant impairment in long-term memory, in an olfactory avoidance conditioning assay.
Flies expressing Ggal\MLCKCaMBP4.Scer\UAS.T:Hsap\MYC under the control of Scer\GAL4c739 for 5 to 6 days prior to testing (using the temperature sensitive Scer\GAL80ts.αTub84B allele to prevent expression during development) do not show defects in short-term memory or in long-term memory in an olfactory avoidance conditioning assay.
