A heat shock promoter drives expression of a kinase-inactive form of "PKMζ". PKMζ is a truncated Mmus\Prkcz isoform which lacks the N-terminal regulatory domain (the mutant protein starts at amino acids 165 of the wild-type protein). The amino acid replacement K281W has been introduced into this isoform, which disrupts kinase activity by altering the ATP-binding domain.
Induction of Mmus\PrkczKI.PKM.hs (using heat shock) either before or after training has no effect on 24 hour memory after single-cycle training in an olfactory avoidance conditioning assay.
Expression of Mmus\PrkczKI.PKM.hs using heat shock prior to training inhibits 24 hour memory after massed training in an olfactory avoidance conditioning assay, while learning (3 hour memory) is not affected.
