Description

UAS regulatory sequences drive expression of a Zzzz\CAG-repeat encoding 55 glutamines. The repeats are flanked by six amino acids on the N-terminal side and four on the C-terminal, and the Tag:MYC and Tag:FLAG epitope tags are located downstream.

Expression of one copy of Zzzz\CAG^{55.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG} under the control of Scer\GAL4^GMR.PFa results in mild eye pigmentation loss, localised mainly to the posterior of the eye. Very few photoreceptors are evident in the ommatidium. The phenotype is more severe when two copies of the transgene are expressed.

Flies expressing Zzzz\CAG^{55.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG} under the control of Scer\GAL4^da.G32 are lethal prior to pupariation. Aggregates are seen in third instar larvae and these are predominantly nuclear.

Brains from adult flies expressing Zzzz\CAG^{55.Scer\UAS.T:Hsap\MYC,T:Zzzz\FLAG} under the control of Scer\GAL4^elav.PLu exhibit a large number of aggregates that are localised to the neuronal cell body. They are found in the central brain, the optic lobes nd the antennae. A single aggregate is seen per nucleus.
