UASt regulatory sequences drive expression of the Hsap\SPAST M87 isoform containing the amino acid replacement C448Y (a pathogenic mutation found in HSP-SPG4 hereditary spastic paraplegia patients).
Flies expressing Hsap\SPASTC448Y.Scer\UAS.M87 under the control of Scer\GAL4elav-C155 do not show any significant postural defect.
The time taken for animals to right themselves is significantly increased compared to wild type in flies expressing Hsap\SPASTC448Y.Scer\UAS.M87 under the control of Scer\GAL4elav-C155. The flies also show significantly reduced climbing ability.
