Description

Imprecise excision of the P{EP}lin-28EP915 progenitor insertion, resulting in 1007bp deletion upstream of the original insertion site (nucleotides 1242-2249 relative to the lin-28 transcription start site are deleted).

Comment:

1007 bp deletion resulting from the imprecise excision of P{EP}lin-28EP915.The deletion is reported at coordinates 1242-2249 relative to the lin-28 transcription start site. Bases on a 2702 nt reported toatl length of the transcript, 3L:5654683 was used as the tx start site. This is two bases downstream from the R6.04 annotated start.

lin-28dF30/lin-28dF30 pupae show significantly reduced volume and weight, but no change in the timing of pupariation compared to wild-type controls.

lin-28Δ1/lin-28dF30 animals start pupariation earlier and have significantly reduced pupal volume and weight compared to wild-type controls, but no change in timing of eclosion from the start of metamorphosis.

Up to 50% of homozygous adults either fail to eclose or die shortly after eclosion.

Homozygous adults show significantly reduced spontaneous and stimulated locomotor activity compared to controls.

Newly eclosed homozygous males (0 to 3 hours after eclosion) show a loss of the transient dorsal internal oblique muscles. 8-day old homozygous males show minor muscle defects, with a shorter than normal muscle of Lawrence and larger than normal dorsal muscles.

Homozygous females show a significantly reduced egg laying compared to controls. 32% of homozygous germaria show an increase in the number undifferentiated germline stem cells

compared to controls. 75.4% of stage 7-10 egg chambers are abnormal, containing either more than the normal 15 nurse cells or showing degeneration. The additional nurse cell phenotype appears to be due to both egg chamber fusion and a mitotic defect (the number of nurse cells seen varies between 22 and 47 and egg chambers containing additional developing oocytes are seen).
