β-PheRS322/Df(3R)Exel6198 (β-PheRS null) flies rescued with the β-PheRSA158W mutant construct have no obvious phenotypes, showing normal developmental time, body size and bristle shape.

β-PheRS322/Df(3R)Exel6198 (β-PheRS null) flies rescued with the β-PheRSA158W mutant construct show normal locomotion (climbing) ability over several weeks. Survival rates also drop after 50 days, compared to controls.

In β-PheRS322/Df(3R)Exel6198 (β-PheRS null) flies rescued with β-PheRSA158W, Scer\GAL4ey.PH-mediated expression of α-PheRSA456G.Scer\UAS results in severe disruption of eye organization - most ommatidia are irregular, the entire eye is smaller and the eye surface is rough. The retina and ommatidia lack regular patterns, shapes and organization because of smaller ommatidia and absence of many cells.

Scer\GAL4Act5C.PI-mediated expression of α-PheRSA456G.Scer\UAS in adults (via use of Scer\GAL80ts.αTub84B) in a β-PheRS322/Df(3R)Exel6198 (β-PheRS null) background rescued with β-PheRSA158W results in many elav-positive brain (neuronal) cells entering apoptosis.

In β-PheRS322/Df(3R)Exel6198 (β-PheRS null) flies rescued with β-PheRSA158W, Scer\GAL4en-e16E-mediated expression of α-PheRSA456G.Scer\UAS results in decreased wing size , and in some wings entire regions of the posterior compartment are missing. In wing discs, the number of apoptotic cells is increased, while cell size is either similar or slightly bigger than controls, suggesting that cell number (proliferation) must be reduced.
