Expression of Act57BK326Q.K328Q.Scer\UAS using Scer\GAL4Mef2.PR predominantly results in pupal lethality with relatively few adult flies emerging from their puparia. The flight ability of the escapers is abolished and they show a characteristic "wings up" phenotype commonly associated with hypercontractile and damaged indirect flight muscles (IFMs). Indeed, polarized light microscopy reveals the absence of continuous IFM fibers.

Scer\GAL4Act88F.PB-mediated expression of Act57BK326Q.K328Q.Scer\UAS causes a complete loss of flight. These flies show prominently hypercontracted IFM fibers, characterized by separation and accumulation of fiber material at one or both attachment sites.
