UASt regulatory sequences drive expression of the C-terminal part of Hsap\WDFY3 (amino acid residues 2285-3526). The protein is tagged at the C-terminal end with EGFP.
Expression of Hsap\WDFY3Cterm.Scer\UAS.T:Avic\GFP-EGFP driven ubiquitously by Scer\GAL4Act.PU does not lead to obvious abnormal phenotypes; when driven by Scer\GAL4GMR.PU there are no obvious eye phenotypes. Compared to expression of Hsap\WDFY3Cterm.Scer\UAS.T:Avic\GFP-EGFP, pharate adults with expression of Hsap\WDFY3R2637W.Cterm.Scer\UAS.T:Avic\GFP-EGFP driven ubiquitously by Scer\GAL4Act.PU have significantly smaller brains (decreased brain volume) that are denser, fragile and malformed; clusters of disorganized cells contain Hsap\WDFY3R2637W.Cterm.Scer\UAS.T:Avic\GFP-EGFP aggregates.
