UAS regulatory sequences drive the highest expression (lethal, line 4) of around 80-100 (0.4-0.5 kb) Zzzz\UGGAA (a repeat tract associated with human SCA31) repeats, followed by a bridging sequence (complex of UGGGAA, UGGAA, UAGAA) and then a (UAGAA)n, (UAAAA UAGAA)n stretch.
This construct encodes only the RNA repeat motif.
The expression of Zzzz\UGGAAL.Scer\UAS under the control of either Scer\GAL4GMR.PS or Scer\GAL4elav-C155 leads to lethality during the pupal stage.
