UAS regulatory sequences drive expression of a constitutively active form of Fic (carries the amino acid replacement E247G). The protein is tagged at the C-terminal end with Tag:V5 and Tag:polyHis.
The expression of FicE247G.Scer\UAS.T:SV5\V5.T:Zzzz\His6 under the control of either Scer\GAL4da.G32 or Scer\GAL4GMR.long in an otherwise wild-type background is viable and adults have no externally visible phenotypes. In an Fic30C homozygosity background, however, Scer\GAL4da.G32-driven expression results in lethality. In Fic30C homozygosity or Fic30C/Df(2L)BSC296 backgrounds, Scer\GAL4GMR.long-driven expression is viable but leads to a small and rough eye phenotype.
The expression of FicE247G.Scer\UAS.T:SV5\V5.T:Zzzz\His6 under the control of Scer\GAL4GMR.long in an Fic30C homozygosity background, leads to a strong increase in apoptosis on the third instar larval eye disc, as compared to the expression of FicE247G.Scer\UAS.T:SV5\V5.T:Zzzz\His6 under the control of Scer\GAL4GMR.long or wild-type controls.
Fic30C homozygotes expressing FicE247G.Scer\UAS.T:SV5\V5.T:Zzzz\His6 under the control of Scer\GAL4GMR.long exhibit a small and rough eye phenotype, which is rescued by the co-expression of FicScer\UAS.T:SV5\V5.T:Zzzz\His6 or FicI271D.Scer\UAS.T:SV5\V5.T:Zzzz\His6, but not FicH375A.Scer\UAS.T:SV5\V5.T:Zzzz\His6.
