UAS regulatory sequences drive expression of Fic carrying the mutations E247G (this mutation results in a constitutively active form of Fic when present by itself) and I271D (this mutation disrupts dimerization). The protein is tagged at the C-terminal end with Tag:V5 and Tag:polyHis.
The expression of FicE247G.I271D.Scer\UAS.T:SV5\V5.T:Zzzz\His6 under the control of Scer\GAL4GMR.long alone is viable and adults have no externally visible phenotypes; in an Fic30C homozygosity background, however, this co-expression leads to lethality.
