The expression of Mmus\Ano6D430G-L.UAS.Tag:FLAG under the control of Ecol\lexAppk.PU leads to dendrite debris from class IV dendritic arborizing neurons being detected on the underlying epidermal cells and leads to a significant decrease in dendrite density and in dendrite field size, as compared to controls.
Ecol\lexAppk.PU, Mmus\Ano6D430G-L.lexAop.Tag:FLAG has larval multidendritic class IV neuron | larval stage phenotype, enhanceable by Spyo\Cas9ppk.PS/Cdc50sgRNA.U6
Ecol\lexAppk.PU, Mmus\Ano6D430G-L.lexAop.Tag:FLAG has dendrite | larval stage phenotype, enhanceable by Spyo\Cas9ppk.PS/Cdc50sgRNA.U6
Ecol\lexAppk.PU, Mmus\Ano6D430G-L.lexAop.Tag:FLAG has dendritic tree | larval stage phenotype, enhanceable by Spyo\Cas9ppk.PS/Cdc50sgRNA.U6
Cdc50sgRNA.U6, Ecol\lexAppk.PU, Mmus\Ano6D430G-L.lexAop.Tag:FLAG, Spyo\Cas9ppk.PS has larval multidendritic class IV neuron | larval stage phenotype, suppressible by drprindel/drprindel
Mmus\Ano6D430G-L.lexAop.Tag:FLAG/Ecol\lexAppk.PU is an enhancer of larval multidendritic class IV neuron | larval stage phenotype of Cdc50sgRNA.U6, Spyo\Cas9ppk.PS
The combined tissue-specific knockout of CDC50 (CDC50sgRNA.U6+Spyo\Cas9ppk.PS) and expression of Mmus\Ano6D430G-L.lexAop.Tag:FLAG under the control of Ecol\lexAppk.PU leads to higher levels of dendrite debris from class IV dendritic arborizing neurons being detected on the underlying epidermal cells than upon either CDC50 knockout or Mmus\Ano6D430G-L.lexAop.Tag:FLAG expression; this phenotype is more severe upon heat shock. This "double mutant" background leads to significant vesicle shedding by class IV dendritic arborizing neurons compared to controls, which is suppressed by drprindel homozygosity.
