Twenty copies of UAS regulatory sequences are separated from Tag:M(mCd8a)-GFP followed by a "T2A" peptide from Thosea asigna, Tag:APEP (a consensus acceptor peptide for biotin ligase, with an additional glutamate residue at the C-terminal end), and the Syt1 coding sequence tagged with mCherry by a FRT flanked transcriptional stop cassette. The T2A peptide ensures that Tag:M(mCd8a)-GFP and mCherry ::Syt1 are produced as separate, individual proteins during translation.
