UASt regulatory sequences drive expression of a mutated form of elgi in which the two cysteines (C18 and C21) in the highly conserved RING domain have been mutated to serines. This results in a protein which cannot auto-ubiquitinate, and which shows dominant-negative activity (interfering with wild-type elgi function). The coding sequence is tagged with Tag:MYC and Tag:HA.
Expression of elgiCS.UAS.Tag:MYC,Tag:HA under the control of Scer\GAL4nub-AC-62 leads to slightly (but significantly) small and rounder wings, a significant decrease in cross vein spacing and a mild hair polarity defect in the proximal wing when compared to controls.
