snRNA:U6:96Ac regulatory sequences drive expression of two sgRNAs (GACTACAGGGATGAGCGAGGAGG and GAATACCACTCCTGCCACCGCGG) that target Trpm. The two sgRNAs are expressed as a single polycistronic tRNA-gRNA cassette.
Germline specific mutagenesis of Trpm, by using TrpmU6:3.gRNA1 as sgRNA source together with Spyo\Cas9nos.PU, results in a significant proportion of females exhibiting rudimentary ovaries. Where ovaries have grossly normal morphology, there is a lower incidence of calcium waves upon in vitro egg activation, as compared to controls. Egg activation still occur in vivo as: unfertilized eggs and embryos are resistant to 50% bleach, which is indicative of vitelline membrane cross-linking; unfertilized embryos still show polar body formation; and early embryos undergo normal mitosis; although fewer larvae hatch.
