Description

The loxP cassette containing a Disc\RFP3xP3.cUa marker has been excised from TI{TI}upCD70-RFP, leaving a single loxP site in its place. The stop codon that was introduced into exon 11 of the endogenous up locus in TI{TI}upCD70-RFP is still present; this is expected to result in a truncated protein that lacks the last 70 amino acids including the C-terminal Glu-rich segment.

Comment:

This allele has an introduced stop codon in exon 11 of up. 57 bases of up sequence are deleted beyond the stop codon and a single loxP site is present in its place. This is expected to result in a truncated up protein missing the last 70 amino acids.

Detailed Description

upCD70/+ adults exhibit decreased average landing height when acute flight ability is assessed, lower endurance when it is tested by repetitive induction of climbing and decreased climbing velocity.

upCD70/+ hearths are more sensitive to to the stress pacing (at 6 Hz for 30 s) while they are more resistant to 5-s episodes of high frequency pacing, with fewer fibrillation and arrest.

upCD70/+ hearths and jump muscles exhibit longer resting sarcomeres. Also upCD70/+ jump muscles exhibit signs of Z-band streaming.
