The RMCE cassette in the Mi{MIC}kibraMI13703 insertion (which is in the kibra fourth intron upstream of exon 5) has been replaced by a sequence cassette consisting of kibra exons 5-9, a T2A site and EGFP tagged with a nuclear localization signal (Tag:NLS(Unk)). A loxP cassette containing a w+* marker has also been inserted, downstream of the T2A-EGFP-Tag:NLS(Unk) sequence. The inserted sequence is expected to be incorporated into the kibra mRNA, recapitulating a mRNA that encodes wild-type kibra protein, while the T2A sequence promotes the separate translation of the EGFP-Tag:NLS(Unk) reporter.
