Genomic DNA has been re-integrated into the attP site in TI{TI}CTCFattP.mCh, reconstituting a CTCF locus that lacks the Cp190 interacting domain (amino acid residues 705-733) and which is tagged at the C-terminal end with three copies of Tag:HA. Some duplicated CTCF sequence (part of exon 3 through to the end of exon 4) is present downstream of the reconstituted locus, but this is not expressed, due to the presence of a SV40 polyadenylation signal between the reconstituted locus and the duplicated sequence. A single loxP site is present in the first intron.
