The Ubi-p63E promoter drives expression of the Hsap\RELA transcription activation domain fused to two copies of 'pMagFast2', a positively charged engineered domain that can heterodimerize with a compatible negatively charged 'nMag'-type domain upon stimulation with blue light (for details of pMagFast2 see PMID:25708714). This protein was designed to act as a hemidriver in the ShineGAL4 system, in which a transcription activation domain fused to a pMag-type domain can combine in vivo with a DNA-binding domain fused to a compatible nMag-type domain upon exposure to blue light, reconstituting a functional transcriptional activator in a light-dependent manner. However, when the Hsap\RELAp65(AD)::pMagFast2.Ubi protein was expressed together with the protein produced by Scer\GAL4DBD.nMagHigh1.Ubi in the presence of blue light, no expression of a reporter was detectable.
