UASt regulatory sequences drive expression of 'CharON' (Caspase and pH Activated Reporter, Fluorescence ON), a reporter that is designed for tracking emerging apoptotic cells and their efferocytic clearance by phagocytes. The CharON reporter consists of two open reading frames (ORFs) separated by a P2A peptide (to ensure separate translation of the two ORFs). The first ORF corresponds to pH-CaspGFP (GC3ai with pH-stabilizing mutation Q204H), a green a fluorescent sensor of caspase activity, the second ORF corresponds to pHlorina (a red fluorescent protein that shows increasing fluorescent with decreasing pH).
