UASt regulates expression of GFP-based reporter of translocation-associated quality control (TAQC). It contains a signal sequence (SS) for co-translational ER targeting, a N-glycosylation site (-CHO) followed by GFP. After that, there is a Tag:FLAG sequence, 20 consecutive lysine residues (K20) known to stall ribosomes during elongation, and mCherry (mCh) downstream of the stalling sequence whose expression indicates translation readthrough. Finally, there is a STOP codon and a polyA sequence.
