UASt sequences regulate expression of membrane-tethered form of nkt in a configuration in which the nkt protein is extracellular, with the N-terminal end of nkt being free. The encoded protein consists of full-length nkt (including the signal peptide and the IgG domain), a Tag:MYC tag that is flanked on both sides by a GN-repeat linker, and the Tag:TM(HSV1-gC) transmembrane domain. The protein is predicted behave as a cell-autonomous peptide ligand that acts on the same cell if a cognate receptor is expressed.
