The genomic region of knrl that is deleted in TI{TI}knrl2xattP (approximately 5.3kb) has been re-integrated into the genome (by replacing the inverted attP site flanked RMCE cassette in TI{TI}knrl2xattP), reconstituting the knrl transcription unit. The re-integrated sequence has 24 copies of the Tag:PP7 stem loop inserted into the first intron. In addition, the promoter-proximal tethering element (coordinates 3L:20620657..20622205 ) has been replaced by an inert sequence (derived from the y gene) of the same length.
