Three copies of the Tag:HA epitope have been introduced at the C-terminal end of the coding sequence (CDS) in the endogenous tkv locus. This allele was generated in a multi-step process that comprised: 1. insertion of sequence containing the last two exons of tkv (and intervening intron), modified to include three copies of the Tag:HA epitope at the C-terminal end of the CDS, into the attP site present in tkvKO.attP, 2. removal of a loxP cassette containing a w+* selectable marker. An attP/B hybrid sequence is present in the last large intron and a single loxP site is also present in the 3'UTR as a remnant of this genome engineering process.
tkv3xHA homozygotes develop normally, display no obvious phenotypic abnormality and are fully fertile.
