Protein complexes were isolated from mitochondria by non-denaturing blue native page. Complex activity was confirmed by in-gel enzymatic staining assays. Bands were excised and subjected to mass spectrometry for protein identification.

COX4, COX5A, COX6B, cype, levy, mt:CoI, mt:CoII were identified as components of mitochondrial OXPHOS Complex IV. COX5A was acetylated.

Source was adult mitochondrial preparation wild-type fly line; proteins produced from endogenous genes.
