A set of transgenic insertion stocks derived by TE mobilization using the Tni\piggyBac-based construct PBac{SAstopDsRed}. The PBac{SAstopDsRed} construct carries a Disc\RFP marker expressed in the eye; to increase insertional mutagenicity, it contains splice acceptors followed by three-frame translational stops in both orientations. The collection consists of second- and third-chromosome lines carrying proximal Scer\FRT sites, to facilitate mosaic analyses.
Mobilization of the PBac construct occurred in animals carrying PBac{SAstopDsRed} on either the X or the second chromosome and a source of Tni\piggyBac\T, piggyBac transposase on II or III. Strains carrying Scer\FRT sites on the second and third chromosomes were used, to facilitate mosaic analysis of the recovered insertional mutations.
Flanking genomic sequences were obtained by sequencing of inverse PCR products, and were mapped by alignment to the genomic sequence.
