    DNA was isolated from adult fly tissue, embedded in agarose blocks and sheared by four freeze/thaw cycles. Ends were polished by treatment with mung bean nuclease and T4 DNA polymerase, and cloned into the pTARBAC6 vector between the BstXI sites.
