Whole flies were ground on dry ice and used for subsequent genomic DNA extraction.
Flies were entrained under 12hr:12hr light:dark cycles for 3-4 days.
Genomic DNA was isolated from whole fly lysate by treatment with SDS, RNase cocktail, proteinaseK and phenol:chloroform extraction. Purified DNA was then sonicated. Sequencing library was constructed per standard Illumina protocol.
