A single-cell transcriptomic study of the diverse populations of cells from the brains of aging flies, from day 0 to day 50 of adulthood.
Reads were processed using Cell Ranger v2.0 (10x Genomics). They were mapped to the Drosophila melanogaster reference genome r6.16. Data from all samples were aggregated using Cell Ranger aggregation pipeline without applying any Cell Ranger normalization method. Further analysis was performed using Seurat. For quality filtering, the scater R package (McCarthy et al., 2017) was used. For dimensionality reduction, 82 principal components were selected based on the 937 most variable genes and used for Shared Nearest-Neighbor (SNN) clustering.
