A characterization of the diverse populations of cells in the adult Drosophila thorax, in wild-type and FoxO and REPTOR overexpression contexts.
The thoraces from thirty adult males were dissected and homogenized. Cells were mechanically dissociated and about ten thousand nuclei per genotype were collected.
Reads were processed using Cell Ranger v7.0.0 (10x Genomics). Further analysis was performed with Seurat v4.3.0. For quality filtering, cells less than 100 UMIs or less than 100 genes were removed. After UMAP dimensionality reduction, clustering analysis was performed with a resolution of 0.3 and clusters were annotated by comparing the top markers with data from the Fly Cell Atlas dataset.
The EMBL-EBI's Single Cell Expression Atlas provides cell-level annotations, clustering data, raw and normalised read counts, and putative marker genes.
