Subject: FlyBase Query (cy548)
Dear Dr White,
I am currently curating your paper for FlyBase:
Koushika et al., 2000, Molec. Cell. Biol. 20(5): 1836--1845
I have a few questions I was hoping you could answer for me.
__elav genomic rescue constructs__
elav genomic rescue constructs are mentioned in a few different places
in your paper, and it is not clear to me how many different constructs
have been used, or whether different names have been given to the same
construct in different places.
In your materials and methods you mention an elav genomic rescue
construct called P{w+=elav+} which was used to make the elavedr
allele. You cite:
Koushika et al., 1996, Curr. Biol. 6(12): 1634--1641
In which the rescue fragment P{elavDmORF} is mentioned. Is this the
construct that is present in the elavedr allele?
Later in your materials and methods you mention a construct you call
P{w+=elav)DmORF2. Is this the same rescue construct as mentioned
above? You cite:
Yao and White, 1991, Molec. Cell. Biol. 11(6): 2994--3000
in which P{elavDmORF} and P{elavDmORF-120} are both mentioned.
elavDmORF
Koushika et al., 1996, Curr. Biol. 6(12): 1634--1641
Samson, 1998, Genetics 150(2): 723--733
Yannoni and White, 1999, J. Cell Sci. 112(24): 4501--4512
Yao and White, 1991, Molec. Cell. Biol. 11(6): 2994--3000
Yao et al., 1993, J. Neurobiol. 24(6): 723--739
Yao and White, 1991, Molec. Cell. Biol. 11(6): 2994--3000
P{elavDmORF}
Provides 72% rescue of lethality of elav5.
Genomic DNA covering 7.5kb of the elav locus, including the ORF,
fused to 800bp of αTub84B trailer sequences.
elavDmORF-120
Yao and White, 1991, Molec. Cell. Biol. 11(6): 2994--3000
P{elavDmORF-120}
Provides comparable rescue of lethality of elav5 to that
provided by elavDmORF.
Deletion of 120 nucleotides (encoding amino acids 35 to 74)
within the region corresponding to the homopolymeric
alanine/glutamine-rich domain.
Is P{w+=elav)DmORF2 one of these? If so does the '2' indicate a specific
insertion?
Thanks in advance for helping me sort this out, I hope to hear from you soon.
Best wishes,
Chihiro
