Subject: FlyBase query
Dear Juan Pablo,
I am curating your paper for FlyBase:
Couso and Bishop, 1998, Int. J. Dev. Biol. 42(3): 345--352
and I have a question about the rn-lacZ construct in Figure 4C and D.
FlyBase doesn't have a record of any rn-lacZ constructs, so I'd be grateful
if you could tell me some details of it.
1. is it an enhancer trap, or a promoter fused directly to lacZ ?
\-- if it is an enhancer trap, what enhancer trap construct is it, eg. lacW,
PZ, and does it have a particular line designation, e.g. 24B, A101.
\-- if the construct is a promoter fused directly to lacZ, how large is the
promoter fragment, what vector is used (e.g. CaSpeR, Carnegie-20), what
marker gene is used, and who made the construct ?
2. are there any references where it has been used ?

Dear Gillian,
The rn-lacZ is an enhancer trap that we found during our screens. It is
a P{lArB}ry+ enhancer trap inserted in the citological location of
rotund, it has a weak rotund phenotype when homozigous (although it is
viable and fertile) and it does not complement rotund alelles. We are
trying to map it molecularly now.

Hi Juan Pablo,
thanks for your e-mail. Do you have a particular number/letter designation
for the rn-lacZ line so that I can record that it was mentioned in the
Int. J. Dev. Biol. paper,
Gillian

We call that rn-lacZ line rn89.
Best regards again,
Juan Pablo.
