Microarray analysis of Df(2R)CX1 Kim Cook, Eric Spana and Kevin Cook DNA samples from Df(2R)CX1 (FBab0022242) heterozygotes were compared to samples from wild type flies by Comparative Genomic Hybridization microarrays at the Duke Model System Genomics Unit as described in Erickson and Spana, 2006 (http://flybase.org/reports/FBrf0193934.html). Corning CGAP slides spotted with the AROS Drosophila V1.1.1 ~70 nucleotide oligo set from Eurofins MWG Operon (www.operon.com) were used for the analysis. Most annotated genes were represented by a single oligo (denoted by a DM number). Sequences present at one copy in deletion heterozygotes are detected by lower relative fluorescence when compared to sequences present in two copies in wild type flies. The left Df(2R)CX1 breakpoint lies within CG12442 or CG18356 or in the region between them, and lies in the range 2R:8587926..8622457 (R5) (predicted cytology: 49C1) based on the following evidence. The gene order at the left Df(2R)CX1 end is CG8768 (FBgn0033769), CG12442 (FBgn0033770), CG18356 (FBgn0033771). A sequence within CG18356 (DM00013773, 2R:8622457..8622525 (R5)) was deleted. A sequence within CG12442 (DM00001050, 2R:8587858..8587926 (R5)) was not deleted, but CG12442 may have been disrupted. A sequence within CG8768 (DM00011116, 2R:8579043..8579111 (R5)) was not deleted. The right Df(2R)CX1 breakpoint lies within Prosap, CG42287 or CG42288 or in a region between them, and lies in the range 2R:9950134..9978645 (R5) (predicted cytology: 50D3-5) based on the following evidence. The gene order at the left Df(2R)CX1 end is CG42287 (FBgn0259183) and CG42288 (FBgn0259184) within introns of Prosap (FBgn0040752), Rcd1 (FBgn0033897). A sequence within Prosap to the left of CG42287 (DM00010589, 2R:9950066 .. 9950134 (R5)) was deleted. A sequence within CG42288 (DM00001813, 2R:9978645..9978713 (R5)) was not deleted, but CG42288 may have been disrupted. A sequence within Rcd1 (DM00010682, 2R:10036111..10036179 (R5)) was not deleted. CG42287 sequences were not present on the microarray. The rest of the microarray data are consistent with genes between CG18356 and Prosap being deleted in Df(2R)CX1.
