The following information accompanied stocks donated to the Bloomington Stock Center by Kent Nybakken, Boston Biomedical Research Institute.
P{UAS-disp.HA.N} was made using a disp cDNA provided by Konrad Basler. The internal Bgl II site in this cDNA was silently mutated to eliminate it. From this modified template, the coding sequence of disp alone (without the stop codon but with the initiator Met) was amplified by PCR with primers providing the disp cDNA with an in-frame N-terminal Bgl II site and an in-frame C-terminal Not I site. Using the Bgl II and Not I sites, this PCR fragment was then cut out and cloned into the Bam HI and Not I sites of a vector made by Kent Nybakken containing an in-frame, C-terminal, triple hemagglutinin (HA) tag followed by a stop codon. The whole triple-HA tagged disp construct was then excised with Bgl II and Xba I, and directionally cloned into the Bgl II and Xba I sites of pP{UAST}. The final arrangement of selected features is Bgl II--Eco RV--disp codons--Not I--triple HA--stop codon--Sma I--Xba I.
P{UAS-disp.HA.N}33 is a second chromosome insertion.
P{UAS-disp.HA.N}79 is a third chromosome insertions.
