The following information accompanied stocks donated to the Bloomington Stock Center by Balaji Iyengar, Hamilton Health Sciences.
M{UAS-PNA.GFP} expresses Peanut Agglutinin (PNA) tagged at the C-terminus with GFP under UAS control.  The sequence for PNA was optimized for Drosophila codon usage and inserted into the SpeI restriction site of a modified version of P{10XUAS-IVS-GFP-WPRE} (pJFRC14; FBtp0056306) in which a SpeI restriction site was introduced upstream of the GFP coding sequence (pJFCR14-CS-dGFP; see http://figshare.com/articles/JFRC14-CS-dGFP.jpg/90134). The plasmid was inserted into the M{3xP3-RFP.attP}ZH-86Fb target site at 86F8,  3R:7634081  (R5) by phiC31 integrase-mediated recombination to generate the M{UAS-PNA.GFP}ZH-86Fb insertion.
M{UAS-WGA2.GFP} expresses Wheat Germ Agglutinin (WGA) tagged at the C-terminus with GFP under UAS control. Drosophila codon-optimized WGA sequence was inserted into the SpeI site of the same vector described above and the construct was inserted into M{3xP3-RFP.attP}ZH-86Fb at 86F8,  3R:7634081  (R5) to generate M{UAS-WGA2.GFP}ZH-86Fb.
Both constructs were made to examine the distribution of glycosylated proteins and their transfer from presynaptic to postsynaptic cells.  See the following online research notes:
