The following information accompanied stocks donated to the Bloomington Drosophila Stock Center by Alexey Pindyurin and Bas van Steensel.
M{hs-dam.4-HT-intein-L127C-HP1} expresses a Dam-Myc-HP1 fusion protein under the control of the full-length hsp70 promoter. The Dam ORF contains the 4-hydroxytamoxifen-inducible intein (Buskirk et al, 2004, PNAS, 101:10505--10510 ) inserted immediately prior to (mutated) position L127C. Insertion of the intein results in reduced activity of the Dam protein.
M{hs.min(FRT.STOP1)dam-HP1} can express a Dam-Myc-HP1 fusion protein (under the control of the minimal hsp70 promoter) after FLP-mediated excision of the FRT-STOP#1-FRT cassette. The STOP#1 transcription termination sequence consists of the C-terminal sequence of the yeast HIS3 gene, the SV40 polyadenylation signal, a false translation initiation signal and a 5' splice donor site.
M{hs-dam.4-HT-intein-L127C-HP1}ZH-51C is an insertion into M{3xP3-RFP.attP'}ZH-51C.
M{hs.min(FRT.STOP1)dam-HP1}ZH-51C is an insertion into M{3xP3-RFP.attP'}ZH-51C.
