NANSDelta23 – A NANS knockout allele generated via CRISPR of Kyoto stk#141875 y* w*; P{FRT(whs)}2A P{neoFRT}82B PBac{SAstopDsRed}LL06554 P{Car20y}96E/TM6B, Tb1. The NANSDelta23 line carries an 893 bp deletion ( 3R:12426793-12427685 ) encompassing a large portion of the two NANS exons. A 20 bp insertion (ATTGTATTGTTGTAACCCAT) of unknown origin (likely inserted during the NHEJ repair process) is found between the deletion endpoints, leading to the conceptual translation of a 78 amino acid long product (MLLNDIISGKLVDSVYIIAEIGQNHQGCVETAKKMIWEAKKAGCHCVKFQKSDLPAKFTRYCIVVTHGLQVAIGRYGH) with the first 60 AAs derived from the endogenous wild-type NANS protein (underlined). The deletion is predicted to eliminate any possible biochemical activity of the NANS protein.
