CG7049R312C - CRISPR/Cas9-mediated homologous dependent repair was used to make amino acid change R312C in CG7049. This change was designed to mimic the R345C change found in human SUMF1 in patients with Multiple Sulfatase Deficiency. The R312C change was put in along with a 3xP3-RFP marker flanked by PBac ends that was then removed via PBac transposition. Transposition leaves behind a TTAA which changes GTCAAC to GTTAAC in a coding region but doesn't change the amino acid sequence.
CG7049S131P - CRISPR/Cas9-mediated homologous dependent repair was used to make amino acid change S131P in CG7049. This change was designed to mimic the S155P change found in human SUMF1 in patients with Multiple Sulfatase Deficiency. The S131P change was put in along with a 3xP3-RFP marker flanked by PBac ends that was then removed via PBac transposition.
TI{TI}CG7049KO - CRISPR/Cas9-mediated homologous dependent repair was used to replace a 904 bp region encompassing most of the CG7049 coding region (from +92 relative to the start ATG to -14 upstream of the stop codon) with a cassette carrying an attP site and lox-P flanked 3xP3-RFP. This results in a null allele of CG7049.
