The strategy for creating the Cep97Δ deletion is as follows and is illustrated in the accompanying figure.
Two Homology arms were cloned into the pGX-attP vector using
1. Not1 and Nde1 for the NT-Frament and
2. Nsi and StuI for the CT-Fragment
We used the following primers
Cep97_KO_pGX_FR1-F(Not1) Ggggacaagcggccgcggcgaaagtcttgtttatttttg
Cep97_KO_pGX_FR1-R(Nde1) Ggggacaacatatgttttctgattggcaggccgaaattc
Cep97_KO_pGX_FR2-F (Nsi) Ggggacaaatgcatacaaatcccagttaataaatg
Cep97_KO_pGX_FR2-R (StuI) Ggggacaaaggccttcgcacattttcatgtgggttc
This resulted in a replacement of the Cep97 CDS with the white gene.
24bp before the start ATG and 54bp before the stop codon.
