The following explains the two associated sequence files. Characterization of the breakpoints of Df(2L)NK01 Genomic DNA from Df(2L)NK01/CyO was used as template for enzymatic amplification with primers OZH-047 and OZH-021. The resulting PCR product (~1.5 kb) was sequenced with the same primers. The sequence obtained with OZH-47 (see file Lehner.2021.7.16-Df2LNK01.gbk) spans the breakpoints. The sequence initially covers a bit of genomic sequences upstream of Nnf1b, runs into residual P element sequence and finally again into genomic sequences from the regions where Dbp21E2 and Saf6 terminate OZH-047: 5’-tcgcaccacaaaaagtcaac-3’ OZH-021: 5’-atatcccggggttagcctggatccttatctttgg-3’ (annealing to genome sequence) Rescue fragment that includes Pex12, Saf6, and Dbp21E2 The sequence of the genomic fragment in the rescue transgene is in the file Lehner.2021.7.16-DSP_region_transgene.gbk. The fly stocks carrying Df(2L)NK01 and/or the gDSP transgene have been discarded in the meantime.
