Ugt37C1 (CG8652) -- I think the first Met in NM_080268.2 should be the start site. Below is the N-terminal alignment of all the 35 Ugt genes from D. melanogaster. Although the first ca. 20 aa are recognized as the signal peptides that are less conserved, Ugt37C1 shows a significant conservation with the other Ugt sequences. If you blast search the deduced amino acid sequence from NM_080268.2 including the first Met, you will get some hits with other Drosophila species such as D. simulans, D. yakuba, D. takahashii, D. biarmipes, D. rhopaloa, etc., although the shorter sequences are also detected in others such as D. sechellia which seems to have a missing 5’-end. From FlyBase (LC): Ugt37C1 (CG8652) -- our current protein annotation is definitely missing amino sequences. Do you have evidence of translation starts at the Met you have indicated? I ask, because it is not conserved. However, the Met 2 bases downstream is conserved and may be the more likely start. The second Met is the start site annotated by the NCBI pipeline (gnomon) for this gene in most of the close Drosophila species.
