TI{TI}Spt5StrepII was generated by insertion of sequences encoding a StrepII tag at the 3’ end of the Spt5 coding region to introduce a C-terminal tag. Sequences for DsRed driven by 3xP3 regulatory sequences were inserted simultaneously. The presence of the tagged Spt5 protein was verified by immunofluorescence staining of polytene chromosomes and western blotting with a StrepII antibody, and by affinity purification of Spt5StrepII from embryo nuclear extract with a StrepTrap XP column.
