M{SuRe.AD1(Bxb1)2} - This SuRe R1 Recombinator construct carries an Adaptor1 (AD1) with a Bxb1 attP site for Bxb1-mediated recombination. This AD1 carries 2xr4-mCherry, a Su(Hw) insulator, a gRNA that targets a specific sequence (AGTGTCGACGATGTAGGTCANGG) adjacent to the 3' UTR and a homology region that recognizes the nanos 3' UTR.
M{SuRe.AD1(Bxb1)2}ZH-2A is an insertion on the X chromosome into ZH-2A (FBti0076477). Use with insertions like M{vas-Bxb1.nanos3'UTR.RFP-}ZH-2A (BDSC#67104), M{nanos-Cas9.P}ZH-2A (w+, RFP+) (BDSC#54591), M{3xP3-GFP.vas-NLS-TP901-1.nanos3'UTR.RFP-}ZH-2A (BDSC#80078), and M{vas-NLS-TP901-1.nanos3'UTR.RFP-.GFP-}ZH-2A (BDSC#605091).

This AD2 carries the marker 2xr4-CFP, a gRNA that targets a specific sequence (CACCGCGCCCAGCGGTTTCGNGG), and a homology region located downstream of the attB site in constructs like M{vas-Bxb1.nanos3'UTR.RFP-}, M{vas-NLS-TP901-1.nos3'UTR.RFP-.GFP-}, M{3xP3-GFP.vas-NLS-TP901-1.nanos3'UTR.RFP-}, and M{vas-Cas9.RFP-}
Note that M{SuRe.AD2(Bxb1)3} cannot be used on M{Act5C-Cas9.P.RFP-} and M{vas-Cas9.RFP-.GFP-} because removing the mini-w+ to create M{Act5C-Cas9.P.RFP-} or GFP to create M{vas-Cas9.RFP-.GFP-} also removed the gRNA target and homologous region.
