The attP cassette of the Mi{MIC} element (in phase 0) has been replaced with a cassette that contains: 1) splice acceptor sequence, 2) PlexB coding sequence after the first exon, 3) a conditional tag (FRT-Tag:FLAG-stop-FRT-Tag:V5-stop), and 4) the 3' UTR of PlexB. In the absence of FLP, PlexB is tagged at the C-terminal with Tag:FLAG. When FLP is expressed the stop sequence is excised, and PlexB is tagged at the C-terminal with Tag:V5.
