Description

20xUAS drives expression of 'green-Camuiα' a fluorescence resonance energy transfer (FRET)-based reporter of CamKII kinase activity. 'green-Camuiα' consists of the entire Rnor\Camk2a coding sequence tagged at the N-terminal end with ReaCh (a non-radiative yellow fluorescent protein variant) and at the C-terminal end with mEGFP. Upon activation, the Rnor\Camk2a protein in green-Camuiα undergoes a conformational change that reduces FRET and increases the fluorescence lifetime of mEGFP; the FRET change reports autophosphorylation at residue T286 and Ca[2+]/calmodulin binding (details from PMID:19295602).
