Description

The FRT stop cassette has been excised, allowing the UAS regulatory sequences to drive expression of the bicistronic sequence that encodes two open reading frames separated by stop codons. The 114bp upstream ORF is simply present to reduce translation of the downstream ORF, which encodes human codon optimized Cas9.
