10xUAS regulatory sequences are followed by an FRT cassette containing four tandem copies of the SNAPf-tag chemical reporter tag, tagged at the N-terminal end with the Tag:Myr(Src64B) membrane targeting sequence (the AcNPV 5'UTR and the P10 3'UTR, shown to increase translational efficiency, are also present). A second open reading frame (ORF) is present after the FRT cassette, encoding 3 tandem copies of the Halo7-tag chemical reporter, tagged at the C-terminal end with the Tag:PM(hKRAS) plasma membrane targeting sequence (the P10 3'UTR, shown to increase translational efficiency, is also present downstream of this second coding sequence). In the absence of FLP recombinase the second ORF is not expressed.
