UAS sequences regulate expression of a fusion protein composed of the hh export signal sequence (codons 1-84), the hh extended N-terminal peptide with a functional palmitoylation acceptor cysteine (codons 85-98), mCherry, amino acids VHG (the three most C-terminal amino acids of the hh signalling domain, the G residue of which is required for cholesteroylation by HhC) and the hh C-terminal domain (HhC, codons 258-472). Autocatalytic cleavage and cholesteroylation by HhC, followed by palmitoylation of the N-terminal acceptor cysteine results in mCherry that is dual-lipidated (with palmitate at the N-terminal end and cholesterol at the C-terminal end).
