The amplitude of glutamate-gated currents is increased in embryos expressing Gad1Scer\UAS.cFa under the control of Scer\GAL4elav.PLu compared to wild-type (2198 +/- 270 pA compared to 1691 +/- 71 pA), when glutamate is pressure-ejected onto the postsynaptic neuromuscular junction. Single-channel current amplitude of postsynaptic glutamate receptors is not different from wild type in the neuromuscular junction of embryos expressing Gad1Scer\UAS.cFa under the control of Scer\GAL4elav.PLu. The amplitude of spontaneous excitatory junctional currents is increased in the neuromuscular junction of embryos expressing Gad1Scer\UAS.cFa under the control of Scer\GAL4elav.PLu compared to wild type (213 +/- 52 pA compared to 141 +/- 14 pA).
Scer\GAL4eve.RKK/Gad1UAS.cFa partially rescues Gad1L352F
The addition of Gad1Scer\UAS.cFa driven by Scer\GAL4elav.PLu to Gad1E143K and Gad1L352F homozygotes, rescues the lethality seen in these flies. 72% of Gad1L352F rescued homozygotes, and 27% of Gad1E143K rescued homozygotes are apparently wild-type.
