Adult flies expressing KankHMS00319 under the control of Scer\GAL4Dot.PK display nephrocyte functional defects (measured by uptake assays) and reduced adult lifespan relative to controls.
Expression of KankHMS00319 under the control of Scer\GAL4Dot.PU in larvae results in impaired nephrocyte function as assayed by fluorescent protein uptake, but no change in number or size of cardiac nephrocytes. Expression of KankHMS00319 under the control of Scer\GAL4Dot.PU results in uneven distribution of endocytic vacuoles and lacuna channels, diminished and abnormally shaped lacuna structures, and a reduced number of vacuoles and lysosomes in cardiac nephrocytes; uneven cell surfaces and gaps between the plasma membrane and the basement membrane of nephrocytes, and absent slit diaphragm filtration structures of the nephrocytes of third instar larvae, as compared to wild type.
Scer\GAL4en-e16E-mediated expression does not significantly change the size of the posterior compartment of the adult wing.
